FOXO4-DRI is a D-amino acid retro-inverso peptide designed to disrupt the FOXO4-p53 protein-protein interaction in senescent cells. Senescent cells accumulate with ageing and drive chronic inflammation (inflammaging) via the senescence-associated secretory phenotype (SASP). By blocking FOXO4 from sequestering p53 in the nucleus, the peptide restores p53's apoptotic function specifically in senescent cells, triggering their elimination without harming healthy cells. Published research in Nature Medicine (Baar et al., 2017) demonstrated reversal of ageing phenotypes and increased physical fitness in mice.
